چکیده

Multimodal T cell profiling can enable more precise characterization of elusive states underlying disease. Here, we integrated single-cell RNA and surface protein data from 500,089 memory cells to define 31 259 individuals in a Peruvian tuberculosis (TB) progression cohort. At immune steady state >4 years after infection disease resolution, found that, accounting for significant effects age, sex, season genetic ancestry on composition, polyfunctional type 17 helper (TH17) cell–like effector was reduced abundance function who previously progressed Mycobacterium (M.tb) active TB These are capable responding M.tb peptides. Deconvoluting this state—uniquely identifiable with multimodal analysis—from public demonstrated that its depletion may precede persist beyond Our study demonstrates the power integrative relevant other traits. Raychaudhuri colleagues use high-dimensional analysis human They identify TH17–like cluster reactive peptides is people

Molecular and Functional Profiling of Memory CD8 T Cell Differentiation

How and when memory T cells form during an immune response are long-standing questions. To better understand memory CD8 T cell development, a time course of gene expression and functional changes in antigen-specific T cells during viral infection was evaluated. The expression of many genes continued to change after viral clearance in accordance with changes in CD8 T cell functional properties. ...

miRNA Profiling of Naïve, Effector and Memory CD8 T Cells

microRNAs have recently emerged as master regulators of gene expression during development and cell differentiation. Although profound changes in gene expression also occur during antigen-induced T cell differentiation, the role of miRNAs in the process is not known. We compared the miRNA expression profiles between antigen-specific naïve, effector and memory CD8+ T cells using 3 different meth...

Associations between Soy Intake and Disease Severity in Cirrhotic Patients: Results from a Cross-Sectional Study

Background and Objectives: Soybean is one of the sources of plant-based proteins and contains various nutrients, including branched chain amino acids, bioactive peptides, isoflavones, soluble fibers and essential omega-3 fatty acids. Furthermore, they include beneficial therapeutic effects on chronic diseases. Target of the current study was investigation of possible associations between soy in...

Fibroblast and T cells conditioned media induce maturation dendritic cell and promote T helper immune response

Dendritic cells (DCs) induce pathogen-specific T cell responses. We comprehensively studied the effects of addition of maturation stimulus, fibroblasts (fibroblast conditioned medium), PHA activated T cells (T cell conditioned medium), and mixture of fibroblast & PHA activated T cells (FCM-TCCM) conditioned media on maturation of DCs. Monocytes were cultured with GM-CSF and IL-4 for five days. ...

Direct ex vivo analysis of antigen-specific IFN-gamma-secreting CD4 T cells in Mycobacterium tuberculosis-infected individuals: associations with clinical disease state and effect of treatment.

The wide spectrum of clinical outcomes following infection with Mycobacterium tuberculosis is largely determined by the host immune response; therefore, we studied several clinically defined groups of individuals (n = 120) that differ in their ability to contain the bacillus. To quantitate M. tuberculosis-specific T cells directly ex vivo, we enumerated IFN-gamma-secreting CD4 T cells specific ...
